The extraction of plant tissue, whether it is root and stem tissue, leaf tissue, or fruit tissue, 

should use fresh weight as the measurement method. Generally, the following principles should be followed:

1.The weight of the tissue to be weighed cannot be less than 50mg.

2.The ratio of homogenate is 10%, that is, 1g of tissue added to 9ml of homogenate solution. The homogenate solution usually selects PBS (pH=7.2-7.4).

3.During the homogenization process, the homogenate solution should be added in 2 times, so that the homogenization is more thorough.

4.After the homogenization is completed, centrifuge the supernatant. The centrifuge speed should be selected from 2000 to 3000 rotations per minute, 
and the speed should not exceed 5000 rotations per minute. The centrifugation time is 15-30 minutes.
