 Hemotropic mycoplasma is parasitic to a variety of In mammalian blood, it is attached to the surface of red blood cells, has no cell wall, is pleomorphic, and can be spread through the blood. Haemophilic mycoplasma was once named as Eperythrozoon and Haemobartonella and was classified as the genus Rickettsia, and has now been reclassified as the genus Mycoplasma based on 16S rRNA gene sequence analysis. Clinically, hemolytic mycoplasma can cause hemolytic anemia. Other symptoms include: anorexia, lethargy, dehydration, weight loss, sudden death, etc. Sometimes it manifests as an acute attack, and many animals have mild or no symptoms after infection. Haemophilic mycoplasma has been found in cats, dogs, pigs, cattle, sheep, camels, rats and other animals, and the blood mycoplasma infection in many animals is still unknown. Haemophilic mycoplasmas infected by different species of animals have genetic similarities, suggesting the possibility of cross-species transmission. Blood-sucking arthropods are generally believed to be the route of transmission.

Haemophilic mycoplasma cannot be cultured in vitro, and the detection method generally uses Giemsa staining Blood smear method. The detection sensitivity and specificity of the blood smear method are relatively poor, and it is easily interfered by the preparation method and other components in the blood. The PCR method is more convenient and more sensitive, and the detection time is short, and the results can be obtained in just a few hours.

This kit performs PCR identification of the 16S rRNA gene, and the primers are specifically targeted. Haemophilic mycoplasma can identify all known haemophilic mycoplasmas that parasitize common mammals ; verified by BLAST, there is no cross-reaction with eukaryotic cell genomes. Cross-reacts only with some uncommon non-haemophilic mycoplasmas, such as: Lionfish Mycoplasma , Mycoplasma fastidiosum, Mycoplasma cavipharyngis pharynx), Mycoplasma insons . Since haemophilic mycoplasma must be extracted from blood, strict operation is required during the sampling process. The above-mentioned cross-reactive mycoplasma can be easily avoided during the sampling process and does not affect the use of the kit. This kit has low sensitivity for wild boar strains of Mycoplasma suis.

100 cat blood DNA samples were tested and 22 positive samples were obtained. Retesting with species-specific PCR can identify the infection as one or more of three types of feline haemophilic Mycoplasma without any false positives. This kit can be used for the identification and detection of haemophilic mycoplasma.

嗜血支原体(Hemotropic mycoplasma,或hemoplasma)寄生于多种哺乳动物血液中,附着于红细胞表面,无细胞壁,具多形性,可通过血液传播。嗜血支原体曾命名为附红细胞体(Eperythrozoon)和血巴尔通体(或血巴通氏体)(Haemobartonella) ,并被分类为立克次氏体属,根据16S rRNA基因序列分析,现已重新归类为支原体属。临床上嗜血支原体可引发溶血性贫血症,其他症状还包括:厌食症、昏睡症、脱水、体重减轻、突然死亡等,有时表现为急性发作,也有很多动物感染后症状轻微或无症状。目前已在猫、狗、猪、牛、羊、驼、鼠等动物中发现嗜血支原体,很多动物的血液支原体感染仍为未知状态。不同种动物感染的嗜血支原体有遗传相似性,提示有跨物种传播的可能性。一般认为吸血性节肢动物是其传播途径。

嗜血支原体尚不能体外培养,检测方法一般采用吉姆萨染色的血涂片法。血涂片法的检测灵敏度和特异性均比较差,容易受到制片方法和血液里其他成分的干扰。而使用PCR法检测则更为方便,且灵敏度更高,检测时间短,仅需几个小时即可获得结果。

本试剂盒针对16S rRNA基因进行PCR鉴定,引物特异性靶向嗜血支原体,可识别寄生于常见哺乳动物(如:猫、犬、猪、牛、羊、驼、鼠等)的所有已知嗜血支原体;经BLAST验证,与真核细胞基因组无交叉反应,仅与一些不常见的非嗜血支原体有交叉反应,如:狮子鱼支原体(Candidatus Mycoplasma liparidae)(发现于深渊狮子鱼肠道)、Mycoplasma fastidiosum(发现于马鼻咽部)、Mycoplasma cavipharyngis(发现于豚鼠鼻咽部)、Mycoplasma insons(发现于绿鬣蜥上呼吸道)。由于嗜血支原体必须从血液中提取,因此只需在取样过程中严格操作,上述有交叉反应的支原体很容易在取样过程中避开,不影响试剂盒的使用。对于猪支原体(Mycoplasma suis)的野猪(wild boar)型菌株,本试剂盒敏感性较低。

对100个猫血DNA样本进行检测,得到22个阳性样本,用物种特异性PCR进行复检,可鉴定出感染的是三种猫嗜血支原体的一种或几种,无假阳性现象。本试剂盒可用于嗜血支原体的鉴定和检测。
