The endoplasmic reticulum is present in various eukaryotic cells except mature red blood cells of mammals. The endoplasmic reticulum is a system of interconnected lamellar gaps or tubules composed of biological membranes. The gap-like spaces between the membranes are called pools, and are usually not directly connected to the extracellular space and the cytoplasmic matrix. This membranous duct system within the cell not only constitutes a pathway for intracellular material transport, but also provides a broad reaction area for various enzyme reactions within the cell. The functions of the endoplasmic reticulum are related to protein synthesis, carbohydrate and lipid synthesis, detoxification, and assimilation, and it also has the function of transporting proteins. Traditional endoplasmic reticulum protein extraction methods require the use of higher centrifugal force or density gradient centrifugation, which requires higher laboratory centrifuge equipment and higher requirements for experimental operation details. If the laboratory does not have equipment that can achieve a centrifugal force of more than 50,000 × g, extraction will not be possible. The endoplasmic reticulum protein extraction kit (low-speed centrifugation method) only requires a centrifugal force of less than 20,000 × g to extract endoplasmic reticulum proteins. General conventional desktop centrifuges can meet the requirements. The endoplasmic reticulum protein extraction kit can be used to extract endoplasmic reticulum proteins from various animal cells and solid soft and hard tissue samples. The protein extracted by this kit can be used for downstream protein research experiments such as Western Blotting, protein electrophoresis, immunoprecipitation, ELISA, transcription activity analysis, Gelshift gel retardation experiment, enzyme activity determination, etc. The proteins extracted by this kit are active proteins with natural protein conformation. This kit does not contain EDTA and is compatible with downstream applications such as metal chelation and chromatography.
