Oxygen free radicals react on lipid unsaturated fatty acids to produce lipid peroxidation. The latter gradually breaks down into a series of complex compounds, including MDA. Lipid oxidation levels can be measured by detecting MDA levels.
MDA was combined with thiobarbituric acid (TBA) to produce red product, which had the maximum absorption peak at 532nm. The content of lipid peroxide in S product could be estimated after colorimetry. At the same time, the absorbance at 600nm was measured, and the content of MDA was calculated by the difference between the absorbance at 532nm and 600nm.
Instruments and reagents required in the test:
Visible spectrophotometer, water bath, table centrifuge, adjustable pipette, 1ml glass cuvette, mortar, ice and distilled water.
Product Content:
Extract: Liquid 60mL×1 bottle, stored at 4℃;
Reagent 1: liquid 30mL×1 bottle, stored at 4℃;
Note: before use, pay attention to whether reagent 1 is completely dissolved. If not, it can be heated at 70℃ and shaken to promote dissolution.
