PubMed PubMed Product Detail Cellular calcium signaling is controlled by the production of inositol phosphates (IPs) by phospholipase C in response to extracellular signals. The IP signaling molecules are inactivated by a family of inositol polyphosphate-5-phosphatases (5-phosphatases). This gene encodes the type II 5-phosphatase. The protein is localized to the cytosol and mitochondria, and associates with membranes through an isoprenyl modification near the C-terminus. Several alternatively spliced transcript variants of this gene have been described, but the full-length nature of some of these variants has not been determined. [provided by RefSeq, Jul 2008]
Function:
Hydrolyzes phosphatidylinositol 4,5-bisphosphate (PtIns(4,5)P2) and the signaling molecule phosphatidylinositol 1,4,5-trisphosphate (PtIns(1,4,5)P3), and thereby modulates cellular signaling events. {ECO:0000269|PubMed:7721860}.
Subunit:
Interacts with APPL1, FAM109A and FAM109B. Interacts with several Rab GTPases, at least RAB1A, RAB2A, RAB5A, RAB6A, RAB8A, RAB9A and RAB33B; these interactions may play a dual role in targeting INPP5B to the specific membranes and stimulating the phosphatase activity. {ECO:0000269|PubMed:17956944, ECO:0000269|PubMed:20133602, ECO:0000269|PubMed:21233288, ECO:0000269|Ref.10}.
Subcellular Location:
Cytoplasm, cytosol. Endoplasmic reticulum-Golgi intermediate compartment. Early endosome membrane. Membrane; Peripheral membrane protein; Cytoplasmic side. Cytoplasmic vesicle, phagosome membrane {ECO:0000250}. Golgi apparatus.
Tissue Specificity:
Platelets.
Post-translational modifications:
Isoprenylation at Cys-990 may be required for localization at the membrane. {ECO:0000269|PubMed:7721860}.
May be proteolytically cleaved after Lys-320 as inferred from N-terminal protein sequence of the 75 kda form. {ECO:0000269|PubMed:1718960}.
Similarity:
Belongs to the inositol-1,4,5-trisphosphate 5-phosphatase type II family.
Contains 1 Rho-GAP domain.
