Chlamydiales, are obligate intracellular parasitic prokaryotes, including 7 major families or familial lineages: Chlamydiaceae, Criblamydiaceae, Parachlamydiaceae, Piscichlamydiaceae, Rhabdochlamydiaceae, Simkaniaceae, Hehua Clinic . Except for Chlamydiaceae, others are generally called Chlamydia-like. Chlamydial microorganisms are pathogenic bacteria in humans and animals. Chlamydia-like parasites are also obligate intracellular parasites, and there are currently few studies. Some of them are also related to human or animal diseases, such as: Parachlamydia acanthamoebae is related to human pneumonia and may also cause cattle. Abortion; Simkania negevensis can cause respiratory tract infections, especially in children; Waddlia chondrophila causes abortion in cattle and probably in humans as well. Some can be worthAmoeba-resistant chlamydiae are also resistant to human macrophages. Therefore, the detection of these chlamydiae and chlamydia-like organisms is of great significance for the prevention and treatment of human and animal diseases.

 

Chlamydia can be detected by culture and serological methods. Since chlamydia is an obligate intracellular parasite, it must be cultured in living tissue or cells, which is complex, time-consuming and has low sensitivity. Serological methods have low sensitivity and are prone to cross-reactivity. Chlamydiae cannot be detected using traditional culture methods. The PCR method has the advantages of high sensitivity, strong specificity, and fast detection speed, which can be completed in only two to three hours, which can effectively make up for the shortcomings of the above two methods. Compared with the ordinary PCR method, the quantitative PCR method can not only accurately quantify, but also be more convenient to operate and less affected by environmental pollution.

 

This kit designs specific primers and probes for the 16S RNA gene. It specifically identifies various microorganisms of the order Chlamydiales. The aforementioned 7 families/lineages cannot be identified only by Piscichlamydiaceae. It has been verified by BLAST that there is no cross-reaction with the genomes of other organisms other than the order Chlamydiales. This kit detects 6 types of chlamydia, 9 types of chlamydia, 9 types of bacteria, and 4 types of amoeba. Only chlamydia and chlamydia-like species produce specific signals, and there is no cross-reaction with other bacteria and amoeba. 422 nasopharyngeal swabs were tested and 48 positive results were obtained, which was consistent with the sequencing results. Piscichlamydiaceae exists in fish. Please note that this kit cannot identify it when using it.

衣原体目(Chlamydiales order ),为专性细胞内寄生的原核生物,包括7个主要的家族或家族性谱系:衣原体科(Chlamydiaceae),Criblamydiaceae,副衣原体科(Parachlamydiaceae), Piscichlamydiaceae, Rhabdochlamydiaceae, 西门坎氏菌科(Simkaniaceae),和华诊体科(或称石德菌科,Waddliaceae)。除了衣原体科,其他笼统称为类衣原体。衣原体科微生物在人或动物中都属于致病菌。类衣原体也属于专性细胞内寄生,目前的研究较少,其中一些成员也与人或动物的疾病有关联,如:Parachlamydia acanthamoebae与人类肺炎有关,还可能在牛引起流产;Simkania negevensis可引起呼吸道感染,尤其是儿童当中;Waddlia chondrophila引起牛的流产,并且很可能在人也同样如此。一些能抵抗阿米巴原虫的类衣原体也同样对人的巨噬细胞有抗性。因此,检测这些衣原体和类衣原体对人类和动物疾病的预防和治疗有重要的意义。

衣原体可以通过培养法和血清学方法进行检测。由于衣原体是专性细胞内寄生,因此必须在活组织或细胞内进行培养,操作复杂,耗时,且灵敏度低。血清学方法的灵敏度较低,且容易产生交叉反应。类衣原体不能采用传统的培养法进行检测。PCR法具有灵敏度高、特异性强的优点,检测速度快,只需两三个小时即可完成,可以有效弥补上述两种方法的不足。定量PCR法与普通PCR法相比,不仅可以精确定量,而且操作更为方便,更少受环境污染的影响。

本试剂盒针对16S RNA基因设计特异性引物和探针,可以特异性识别衣原体目的各种微生物,前述7种家族/谱系仅Piscichlamydiaceae不能识别,经BLAST验证,与衣原体目以外的其他生物基因组没有交叉反应。本试剂盒检测了6种衣原体,9种类衣原体,9种细菌,以及4种阿米巴原虫,只有衣原体和类衣原体产生特异性信号,与其他细菌和阿米巴原虫没有交叉反应。对422个鼻咽拭子进行检测,获得48个阳性,与测序结果一致。Piscichlamydiaceae存在于鱼类,使用时需注意本试剂盒不能识别。
