Chlamydiales order belongs to the broad sense of bacteria, which are Gram-negative spherical microorganisms that are strictly intracellular parasitic and have unique development cycles, including 7 Major families or familial lineages: Chlamydiaceae, Criblamydiaceae, Parachlamydiaceae, Piscichlamydiaceae em>, Rhabdochlamydiaceae, Simkaniaceae, and Waddliaceae . With the exception of Chlamydiaceae, other members are collectively referred to as chlamydoids. Chlamydial microorganisms are pathogenic bacteria in humans and animals. Chlamydia-like organisms are also obligate intracellular parasites, and there are currently few studies. Some of them are related to human or animal diseases, such as: Parachlamydia acanthamoebae is related to human pneumonia and may also cause abortion in cattle. ; Simkania negevensis can cause respiratory tract infections, especially in children; Waddlia chondrophila causes abortion in cattle and probably in humans as well. Some chlamydial species that are resistant to amoeba are also resistant to human macrophages. Therefore, the detection of these chlamydiae and chlamydia-like organisms is of great significance for the prevention and treatment of human and animal diseases.

 

Chlamydia can be detected by culture and serological methods. Since chlamydia is an obligate intracellular parasite, it must be cultured in living tissue or cells, which is complex, time-consuming and has low sensitivity. Serological methods have low sensitivity and are prone to cross-reactivity. The PCR method has the advantages of high sensitivity, strong specificity, and fast detection speed, which can be completed in only two to three hours, which can effectively make up for the shortcomings of the above two methods.

 

This kit designs primers for the 16S rRNA gene interval sequence and specifically identifies all members of the order Chlamydiales. It has been verified by BLAST that it is compatible with other organisms other than the order Chlamydiales. There is no cross-reactivity in the genome. This kit tested 64 pig samples and obtained 14 positive results. Sequencing confirmed that 10 of them were Chlamydia suis and 1 was similar to Chlamydia psittaci, and 3 are unknown chlamydia.

衣原体可以通过培养法和血清学方法进行检测。由于衣原体是专性细胞内寄生,因此必须在活组织或细胞内进行培养,操作复杂,耗时,灵敏度低。血清学方法的灵敏度较低,且容易产生交叉反应。PCR法具有灵敏度高、特异性强的优点,检测速度快,只需两三个小时即可完成,可以有效弥补上述两种方法的不足。

本试剂盒针对16S rRNA基因区间序列设计引物,特异性识别衣原体目所有成员,经BLAST验证,与衣原体目以外的其他生物基因组没有交叉反应。本试剂盒检测了64个猪样本,获得14个阳性,测序证实其中10个为猪衣原体(Chlamydia suis),1个类似鹦鹉热衣原体(Chlamydophila psittaci),还有3个属于未知衣原体。
