Chlamydia felis, also known as Chlamydia felis/Chlamydia felis, is a specialized Sexual intracellular parasitic prokaryote, Gram-negative, belonging to the family Chlamydiaceae, once classified as a subtype of Chlamydophila psittaci in cats, was officially established as an independent species in 1999. It is generally believed that the host of Chlamydia felis is limited to cats, but it is also found in dogs and may also cause keratoconjunctivitis in humans; Chlamydia felis cannot survive after leaving the host, so it must be transmitted through close contact , young cats are more susceptible to infection. Feline chlamydia is a common infection and the main cause of feline conjunctivitis. The eye symptoms include conjunctivitis first on one side and then on both sides, nictitating membrane congestion, blepharospasm, increased eye secretions, and conjunctival edema. In addition, it can also cause Fever, lethargy, lameness, weight loss, and some upper respiratory symptoms. Chlamydia felis can be cultured in chicken embryos or cell lines. For the detection of feline chlamydia, the culture method is more difficult because the conjunctival swab contains only a small amount of bacteria, and the antibodies in the tears may also have an impact during chronic infection; and the culture method is time-consuming and complicated to operate. . The PCR method is an in vitro enzymatic synthesis method of specific DNA fragments. It is more suitable for detecting feline chlamydia. Its sensitivity is higher than the culture method, and it has strong specificity and fast detection speed, which can be completed in only two or three hours. Compared with the ordinary PCR method, the quantitative PCR method can not only accurately quantify, but also be more convenient to operate and less affected by environmental pollution.

 

This kit designs primers and probes for the extracellular membrane protein gene sequence. It can specifically identify Chlamydia felis and has been verified by BLAST to have no cross-reactivity with other biological genomes. This kit detected 5 other chlamydia and 14 types of bacteria, and no non-specific signals were found; 272 samples from 9 domestic animals were tested and 139 positive results were obtained, of which 135 were from cats and 4 were from affected conjunctiva. 猫衣原体可以在鸡胚或细胞系中进行培养。对猫衣原体的检测,培养法较为困难,因为结膜拭子中只含有很少量的菌体,并且在慢性感染时眼泪中的抗体也会造成影响;而且培养法较为耗时,操作也比较复杂。PCR法是一种体外酶促合成特异性DNA 片段的方法,更适合检测猫衣原体,其灵敏度高于培养法,且特异性强,检测速度快,只需两三个小时即可完成。定量PCR法与普通PCR法相比,不仅可以精确定量,而且操作更为方便,更少受环境污染的影响。

本试剂盒针对胞膜外蛋白基因序列设计引物和探针,可以特异性识别猫衣原体,经BLAST验证,与其他生物基因组没有交叉反应。本试剂盒检测了5种其他衣原体和14种细菌,未发现非特异性信号;对源自9种家养动物的272个样本进行检测,获得139个阳性,其中135个来自猫,4个来自患结膜炎的狗。可见本试剂盒适用于猫衣原体的检测和鉴定。
