Mycoplasma hyovis is present in the tonsils and upper respiratory tract of many pigs, and these pigs may become lifelong carriers. Mycoplasma suis synovialis can circulate in the blood and has an affinity for joint tissue, infecting joints and tendon sheaths, causing non-suppurative arthritis, resulting in obvious lameness and pain. It is easy to cause disease in growing pigs aged 8-30 weeks, but it is very rare in older sows because the immune resistance of the pig body has already formed. This immune resistance can be passed to offspring through colostrum. and gradually disappear over several weeks.

 

Both in vitro culture method and PCR method can be used to detect Mycoplasma in porcine synovial fluid. In vitro culture method is a classic detection method, but it takes a long time. The use of PCR method has higher sensitivity and short detection time, and the results can be obtained in only a few hours.

Porcine synovial fluid Mycoplasma PCR detection kit selects a sequence of the 16S rRNA gene PCR identification was performed, and the primers were verified by BLAST to specifically target Mycoplasma porcine synoviae and had no cross-reaction with the genomes of other organisms. This kit was used to detect 724 samples obtained from 270 pigs, and only Mycoplasma porcine synoviae produced specific amplification bands. 14 positive samples were detected out of 724 samples, among which the positive rate of tonsils was 4.14%, the positive rate of synovial fluid was 1.60%, and the positive rate of lung tissue was 0. It can be seen that this kit is species-specific and can be used for the identification and detection of Mycoplasma in porcine synovial fluid.
