Mycoplasma is a type of deficiency Prokaryotes with cell walls generally range in size from 0.3 to 0.5um and are highly pleomorphic, with various shapes such as spheres, rods, filaments, and branches. It is different from bacteria and viruses. It has many types and is widely distributed. It causes considerable harm and involves many fields such as humans, animals, plants and insects. Mycoplasma bovis is the causative agent of bovine infectious mycoplasma pneumonia . It is the most important pathogenic bovine parasitic mycoplasma and has a global epidemic trend. It can cause bovine pneumonia, Various diseases such as arthritis, mastitis, reproductive disorders and miscarriage have an incidence rate of 60%-100%, but the mortality rate is not high. Mycoplasma bovis does not always cause disease in cattle and can be found in both healthy and diseased cattle. M. bovis is difficult to diagnose and control because its disease manifestations and response to treatments and vaccines often vary.
The detection of Mycoplasma bovis can be done through serological methods, or genetic methods, for detection. Later studies found that there were antigenic and genetic variations between laboratory strains of Mycoplasma bovis and strains isolated in the field, which interfered with the above detection methods.
PCR based on stable housekeeping genes can eliminate the interference of the above mutations. Therefore, the Mycoplasma bovis PCR detection kit selected a gene related to DNA repair with very small intraspecific variation for PCR identification. The primers were verified by BLAST to specifically target Mycoplasma bovis and do not cross-react with the genomes of other organisms. This kit was used to detect 22 different bovine or ovine parasitic mycoplasmas, and only Mycoplasma bovis produced specific amplification bands. This kit can produce specific amplification bands for 13 Mycoplasma bovis strains isolated in laboratories or in the field. It can be seen that this kit is species-specific and can be used for the identification and detection of Mycoplasma bovis.

支原体是一类缺乏细胞壁的原核生物,大小一般在0.3~0.5um之间,呈高度多形性,有球形、杆形、丝状,分枝状等多种态。它不同于细菌,也不同于病毒,种类繁多,分布广泛,造成的危害相当大,涉及人、动物、植物及昆虫等多个领域。牛支原体(Mycoplasma Bovis)是牛传染性支原体肺炎(又称牛烂肺病)的病原体,是最重要的致病性牛寄生支原体,呈全球流行趋势,可引起牛肺炎、关节炎、乳腺炎、生殖紊乱和流产等多种疾病,发病率达到60%-100%,但病死率不高。牛支原体并不总是引起牛的疾病,在健康或患病的牛身上均可发现其存在。由于牛支原体的疾病表现和对治疗和疫苗的反应常发生变化,对它的诊断和控制较为困难。
对牛支原体的检测,可以通过血清学方法(如免疫荧光法和免疫印迹法),或遗传学方法(如基于RRS基因的PCR法或寡核苷酸互补配对), 进行检测。后来的研究发现牛支原体的实验室菌株和野外分离获得的菌株存在抗原和遗传学上的变异,对上述检测方法形成了干扰。
基于稳定的管家基因的PCR可以排除上述变异的干扰。因此,牛支原体PCR检测试剂盒选取了一个种内变异很小的与DNA修复有关的基因进行PCR鉴定,引物经BLAST验证为特异性靶向牛支原体,与其他生物的基因组不产生交叉反应。使用本试剂盒检测了22种不同的牛或羊寄生性支原体,仅有牛支原体产生特异性扩增条带。而对13个实验室或野外分离的牛支原体菌株,本试剂盒均可产生特异性扩增条带。可见本试剂盒具有物种特异性,可用于牛支原体的鉴定和检测。
