山羊肺炎支原体在体外生长较慢,较难培养,其菌落与丝状支原体族其他成员不易区分,在血清学检测上也常有交叉反应。因此,使用PCR法进行检测,既可以达到很高的灵敏度,也可以避免其它支原体带来的干扰。
山羊肺炎支原体PCR检测试剂盒选取了基因组内与无氧代谢有关的一段序列进行PCR鉴定,引物经BLAST验证为特异性靶向山羊肺炎支原体,与其他生物的基因组不产生交叉反应。使用本试剂盒检测了14种山羊肺炎支原体菌株,以及27种血缘关系较近的其他丝状支原体菌株,仅有山羊肺炎支原体产生扩增条带(316bp)。该条带可被核酸内切酶ClaI切割为190和126 bp两个片段。即使在与其他菌种(包括其他丝状支原体菌株)混合的情况下,本试剂盒也可特异性检测出山羊肺炎支原体,可见本试剂盒具有物种特异性,可用于山羊肺炎支原体的鉴定和检测。
